Introduction: Adult Mesenchymal stem cells (MSCs), showing a high capacity of multipotency and differentiation into various tissues, have been primarily used for the biological repair of cartilage and bone. MSCs can be isolated from various organ- tissues and Periosteum seems to be an excellent source of MSCs in bone engineering.

Hypothesis: We hypothesized that Periosteum derived MSCs present similar surface marker profile to Bone Marrow derived MSCs, adipose derived MSC and sponge bone derived cells, and express positive cell osteogenic markers, indicating potential osteoblastic differentiation.

Material and Methods: We isolated and cultured cells from all three sources; bone marrow (BM), adipose tissue (AT), periosteum (PO), and we used osteoblast-like cells from spongy bone (SB) as positive control. Morphology of cells was analyzed by light microscopy and cytoskeleton by  immunofluorescence staining. The cells were characterized by flow cytometry for surface markers which are well established for MSCs characterization. Cells were also analyzed by immunohistochemistry for the expression of osteogenic markers; alkaline phosphatase, collagen I, and osteopontin.

Results: PO-MSCs demonstrated positive markers for MSCs and lack of markers for hematopoietic cells. Also PO-MSCs had similar properties to BM-MSCs and to our positive control, SB-Cs.Cells derived from BM, PO-MSCs were positive for osteogenic markers; alkaline phosphatase, collagen I, and osteopontin. Periosteum derived cells presented minor differences in morphology and cytoskeleton as revealed by Phalloidin staining,

Conculsion: All these findings suggest that periosteum could be used as a good source of MSCs, described as PO-MSCs, which demonstrate similar morphology, surface marker profile, and osteogenic properties to bone marrow, and adipose derived MSCs.
