Polyfenoly predstavujú rozmanitú skupinu prirodzene sa vyskytujúcich látok, ktoré sa vo vedeckej oblasti tešia nemalej pozornosti. Hlavným dôvodom záujmu o hlbšie poznanie polyfenolov je ich nezameniteľný biologický potenciál, ktorý zahŕňa frapantné zdravotné benefity pre konzumenta. Ochrana pred oxidačným stresom, poškodením buniek ako aj prevencia kardiovaskulárnych ochorení zvýrazňujú dôležitosť konzumácie týchto prírodných látok. Hlavným cieľom tejto práce bolo posúdenie vplyvu polyfenolov na vybrané hormóny v krvi subjektov mužského pohlavia. V štúdii participovalo 48 probandov s vekovým priemerom 30,21 +/- 8,9 rokov. Odber krvných vzoriek podstupovali probandi na lačno, pričom sa krv odoberala do skúmaviek Monovette. Krvné sérum sme získali centrifugáciou pri 3000 RPM po dobu 10 minút pri laboratórnej teplote. Následne bolo sérum uskladnené pri -80 °C do doby vykonania analýz. Odber vzoriek prebiehal v 0. týždni ( Kontrola I.), následne v 3. , 6. a 12. týždni, pričom po uplynutí 24 týždňov prebehol finálny odber ( Kontrola II.). Počas trvania odberových týždňov ( 3,6,12 ) konzumovali subjekty výživové doplnky s obsahom polyfenolov, v dávkach prispôsobených telesnej hmotnosti jednotlivca. Hlavnými zložkami podávanej výživy boli kvercetín a epikatechín. Cieľom nami vykonaných analýz bolo sledovať zmenu hladiny hormónov: androstendión (ANDRO), dehydroepiandrosterón – sulfát (DHEA-S), folikulostimulačný hormón (FSH), luteinizačný hormón (LH), testosterón (TEST) a  prostatický špecifický antigén (PSA) pred, počas a po konzumácii polyfenolických výživových doplnkov stravy. Analýzy sme vykonali za použitia metódy ELISA. Merania prebehli za použitia spektro-fluoro-luminometra (GloMax®Multi+ DetectionSystem, Promega) pri 450nm. Po vykonaní štatistickej analýzy môžeme konštatovať medzi sledovanými hormónmi štatisticky nesignifikantné rozdiely (P> 0,05). Pri sledovaní obsahu glykoproteínu PSA sme zaznamenali pokles hladiny v priebehu konzumácie polyfenolov, čo môžeme považovať za klesajúci trend v časovom horizonte užívania, pričom tento pokles len potvrdzuje zdravotné benefity pri konzumácii. Analýza obsahu DHEA-S preukázala mierny nárast tohto steroidného hormónu, čo môže byť dôsledkom veku probandov. Mierne zvýšenie hodnôt sme zaznamenali taktiež u ostatných spomínaných hormónov (TEST, FSH, LH, ANDRO). Zo získaných dát môžeme vyvodiť jasný záver, napriek štatisticky nesignifikantným rozdielom v hladinách hormónov v nami použitom časovom horizonte, môžeme predpokladať signifikantnejšie rozdiely v dlhšom časovom pásme užívania. Dôvodom predpokladu je najmä mierne stúpajúci či klesajúci trend hladín hormónov.

Polyphenols represent a diverse group of naturally occurring substances that enjoy considerable attention in the scientific field. The main reason for interest in deeper knowledge of polyphenols is their unmistakable biological potencial, which includes frapant health benefits for the consumer. Protecion against oxidative stress, cell damage as well as prevention of cardiovascular diseases highlights the importance of consuming these natural substances. The main objective of this work was to evaluate the impact of polyphenols on selected hormones in the blood of male subjects. The study involved 48 male subjects with an age average of 30.21 +/- 8.9 years. Blood samples were taken from empty stomach subjects to Monovette tubes. We obtained the blood serum by centrifugation at 3000 RPM for 10 minutes at room temperature. Subsequently, the serum was stored at -80°C until implemented analyses. Sampling took place in week 0 (Control I.), followed by week 3., 6. and 12., with final sampling after 24 weeks (Control II.). During the sampling weeks (3,6,12) subjects consumed nutritional supplements containing polyphenols, in doses adapted to the individual´s body weight. The main components of the served nutririon were quercetin and epicatechin. The aim of our analyses was to monitor the change in levels of hormones: androstenendione (ANDRO), dehydroepiandrosterone-sulphate (DHEA-S), follicle-stimulating hormone (FSH), luteinizing hormone (LH), testosterone (TEST) and prostate specific antigen (PSA) before, during and after consumption of polyphenolic dietary suplements. Analyses were carried out by the ELISA method. Measurements were made using a spectro-fluoro-luminometer (GloMax®-Multi+ DetectionSystem Promega) at 450nm. After conducting a statistical analysis, we noted statistically non-significant differences (P> 0.05), between observed hormones. When monitoring the level of glycoprotein PSA, we observed a decrease in the level during polyphenol consumption, which can be considered a downward trend in the time horizon of use, this decrease only confirms the health benefits of polyphenol consumption. Analysis of the content of DHEA-S showed a slight increase in this steroid hormone, which may be due to the age of the probands. We also experienced a slight increase in values in other mentioned hormones (TEST, FSH, LH, ANDRO). From the data obtained we can draw a clear conclusion, despite statistically non-significant differences in hormone levels in the time horizon used by us, we can assume more significant differences in the longer time zone . We made this assumption  mainly due to a slightly increasing or decreasing trend in hormone levels during the consumption time.

Hello. It is a very interesting study. What was the polyphenol concentration in these supplements? In my research, I deal with the design of plant-based beverages made from legumes and the determination of their polyphenol content, so I wonder if their regular consumption could also affect certain hormones in the blood.

Hello Patrycja, thank you very much. You may find concentration of polyphenols in my presentation (9th slide), subjects had to ingest polyphenol dose due to their body weight. For example - 75kg man had to eat 8 capsules per day (the more you weigh, the more capsules you have to eat). 1 capsule weights 0,598g +/- 5%. With the table in my presentation, you can use ratio (μg/g) to compute exact concentration of polyphenols in 1 capsule,as well as in whole dose. Of course legumes polyphenols can also enrich fluctuation of hormone levels, are you planning to examine effect of legumes on hormone levels in your next studies ?
